人脑星形细胞瘤组织中转化生长因子α与肿瘤增殖细胞核抗原检测

摘要 中图分类号R739.41摘要目的:探讨人脑星形细胞瘤组织中转化生长因子α(TGFα)蛋白的表达与肿瘤增殖细胞核抗原(PCNA)及预后的关系。方法:采用免疫组化SP法检测50例人脑星形细胞瘤(其中星形细胞瘤18例,间变性星形细胞瘤20例,多形性胶质母细胞瘤12例)石蜡标本和10例正常脑组织中TGFα蛋白表达和PCNA标记指数(PCNALI),并随访比较50例人脑星形细胞瘤患者术后1a的生存率。结果:10例正常脑组织中TGFα、PCNALI蛋白无表达;50例人脑星形细胞瘤组织中TGFα蛋白表达总阳性率为60.0%(30/50),总PCNALI为(67.86±28.68)%;随着肿瘤病理分级的增高,TGFα蛋白表达和PCNALI也显著增高,各级别组间差异有统计学意义(P<0.05);TGFα蛋白表达和PCNALI呈正相关(r=0.864,P<0.01),TGFα蛋白表达强弱和PCNALI高低反映患者预后(P<0.01)。结论:人脑星形细胞瘤中存在TGFα和PCNA蛋白高表达,联合检测TGFα蛋白和PCNALI对评估人脑星形细胞瘤的生物学行为具有重要意义。 Aim : To study the expression of transforming growth factor α(TGF-α) and proliferating cell nuclear antigen(PCNA) in human astrocytoma tissue. Methods:The expression of TGF-α and PCNA protein among 50 surgical specimens of human astrocytoma and 10 cases of normal brain tissue was detected using immunohistochemical staining method. All patients were followed up. Results: The total positive rates of TGF-α protein was 60.0% (30/50) , and the total PC- NA-LI was 67.86% ±28.68% in 50 surgical specimens of human astrocytoma. The expression of TGF-α and PCNA-LI increased with the increase of pathological grade ( P 〈 0. 05 ). There was positive correlation between TGF-α and PCNA-LI (r =0. 864 ,P 〈0. 01 ). The expression of TGF-α and PCNA protein was related to the patients'prognosis (P 〈 0.01 ). Conclusion : There are overexpressions of TGF-α and PCNA protein in human astrocytoma tissue. Combined examination of TGF-α and PCNA protein has significance in evaluating the biological behavior of human astrocytoma.
