摘要
目的:探讨糖尿病大鼠血糖水平与视网膜微血管中糖基化终产物受体(RAGE)、细胞间黏附分子-1(ICAM-1)表达及无细胞毛细血管数的关系.方法:复制链脲佐菌素(strep-tozoc in,STZ)糖尿病大鼠模型,成模后每4 wk监测血糖,采用免疫组织化学方法和半定量方法分析视网膜微血管中RAGE,ICAM-1的表达,光学显微镜观察视网膜微血管形态学改变.结果:糖尿病大鼠视网膜微血管中RAGE,ICAM-1的表达较非糖尿病组分别增加1.27,1.41倍,无细胞毛细血管数增加2.68倍(P<0.05),与血糖升高成线性相关(r分别为-0.693,-0.805,0.855,P<0.01).结论:糖尿病大鼠视网膜中无细胞毛细血管,RAGE及ICAM-1表达的增加与血糖水平密切相关.
AIM: To study the relationship of the blood glucose of diabetic rats with the expressions of receptor for advanced glycation end products (RAGE), intercellular cell adhesion molecule-1 (ICAM-1) and the number of acellular capillaries. METHODS: Animal models were induced by streptozocin (STZ). Blood glucose levels were monitored every 4 weeks after the induction of diabetes. The accumulation of the immunoreactive RAGE and ICAM-1 were monitored and analyzed by the immunohistochemical assessment and the semi-quantitative analysis. The morphological changes of retinal microvasculature were examined with a light microscope. RESULTS: The immunoreactivities of RAGE and ICAM-1 of diabetic group (Db) increased 1.27-fold and 1.41-fold compared with those of none diabetic group (Non-Db). Acellular capillaries in Db group increased 2.68-fold ( P 〈 0. 05 ). Blood glucose had a significant correlation with the immunoreactivities of RAGE (r = - 0. 693, P 〈 0. 01 ) and ICAM-1 (r= -0.805, P〈0.01) and the number ofacellular capillaries (r = 0. 855, P 〈 0. 01 ). CONCLUSION: The increases of immunoreactivities of RAGE, ICAM-1 and the number of acellular capillaries in diabetic rats are correlated with the level of blood glucose.
