噻唑蓝比色法测定化学物皮肤致敏性的研究 被引量:5

摘要 目的:研究噻唑蓝比色法在检测和评价化学物皮肤接触致敏性中的应用。方法:将致敏物质肉桂醛、DNCB和化妆品纤体精华液、防晒型营养水分别对BALB/c小鼠双耳进行连续3 d染毒,第5天取小鼠耳部淋巴结制备单细胞悬液,用噻唑蓝比色法检测淋巴细胞增殖,用方差分析进行比较。同时采用传统实验方法-局部封闭涂皮法对DNCB、纤体精华液与防晒型营养水进行皮肤变态反应试验。结果:与溶剂对照组比较,肉桂醛与DNCB能引起淋巴细胞的显著增殖,与空白对照组比较,纤体精华液与防晒型营养水未引起淋巴细胞明显增殖,经传统实验方法检测证实,DNCB引起75%动物皮肤变态反应,而纤体精华液与防晒型营养水未引起动物皮肤变态反应。结论:噻唑蓝比色法具有评估化学物皮肤接触致敏性的能力,对化妆品皮肤致敏性评价具有较好的应用前景。
