胆囊癌细胞运动侵袭相关基因的筛选

Identification of motility and invasion related genes in gallbladder cancer cell lines by microarray

摘要 目的筛选与胆囊癌(GBC)细胞运动、侵袭等生物学行为密切相关的基因,探讨胆囊癌转移的分子生物学机制。方法微阵列技术检测两个分别来源于肿瘤原发灶和转移灶,并具有不同运动侵袭能力的胆囊癌细胞系全基因表达的差异。结果基因芯片中待检的15类5832个基因中。2倍以上表达差异基因共261个,去除分类重复,上调基因78个,其中3倍以上差异17个;下调基因69个,其中3倍以上差异14个。应用逆转录-聚合酶链反应(RT-PCR)验证部分基因的表达差异。证实基因芯片的结果可靠。结论肿瘤的转移是多基因异常改变的结果。以上结果经过进一步的实验验证后可能是胆囊癌进展过程中的关键因子,也有可能成为胆囊癌治疗的靶基因。 Objective To screen out motility and invasion related genes in gallbladder cancer (GBL) and explore the molecular biological mechanism of GBL. Methods Two GBC cell lines T1 and T2 from the same tumor but different original sites, having different motility and invasion ability, were detected for the difference in the expression of genes by microarray. Results In the tested 5 832 genes from different categories in the genetic microchips, 261 genes had the alteration of transcription level. Eighty-one genes had more than 2 times higer transcription level in T2, 17 3 times higher. 69 genes transcript in 2 times lower level in T2, 14 have 3 times lower level. Two genes were selected for further reverse transcriptase-PCR analysis. We concluded that the result of microarry was credible. Conclusion The metastasis of GBC concerns a great deal of genetic alternations in a complex network, The result of microarray provides some clues for further investigation in metastasis mechanisms of GBC.
