摘要
目的:以逆转录(RT)和实时定量聚合酶链式反应(Real-tim e qPCR)检测和分析药物对于非酒精性脂肪肝病(NAFLD)干预效果。方法:以高脂饲料饲养W istar大鼠12周,诱发NAFLD,然后转以基础饲料饲养,并对实验大鼠进行多烯磷脂酰胆碱干预与蒸馏水对照的实验,每组大鼠8只,为期4周。提取脂肪组织的总RNA,逆转录为cDNA,通过实时定量PCR检测肿瘤坏死因子α(TNF-α)mRNA的相对水平,并以t-检验进行数据的统计分析。结果:干预组大鼠脂肪组织TNF-αmRNA的相对水平(0.269±0.047)高于对照组(0.125±0.015),两组间差异显著(t=3.094;P=0.009)。结论:多烯磷脂酰胆碱干预可显著增加NAFLD大鼠脂肪组织TNF-α基因的表达而不利于实验大鼠肝内的糖代谢过程。
Objective: To detect and analyze the effects of medical intervention for non-alcoholic fatty liver disease (NAFLD) using reverse transcription (RT) and real-time quantitative polymerase chain reaction (real-time qPCR). Methods: The Wistar rats were fed with high fatty diet for 12 weeks to induce NAFLD, and then were switched to low fatty diet, intervened with polyene phosphatidylcholine, compared with controls receiving distill water for 4 weeks. Each group consisted of 8 rats, The total RNA in adipose tissue was extracted and reversely translated into cDNA for detection of the relative level of tumor necrosis factor a (TNF-a) using real-time qPCR. T-test was performed for statistical significance of the data obtained. Results: The relative level of TNF-α mRNA in adipose tissue of intervention group (0,269 ±0,047) were higher than that of control group (0, 125 ±0,015), The difference of TNF-α mRNA between the two groups was significant ( t = 3. 094 ; P = 0. 009), Conclusion: Polyene phosphatidylcholine can significantly increase the expression of TNF-α gene in adipose tissue of NAFLD rats undermining glucose metabolism in liver.
