摘要
目的:探讨血小板衍生生长因子-BB(PDGF-BB)对人牙周膜(PDL)细胞在离体牙根面上附着、增殖的影响。方法:应用细胞计数法和扫描电镜观察法,观察在细胞因子作用下,离体正常和牙周病患牙根面上PDL细胞附着和增殖的情况。结果:无论是否加入PDGF-BB,PDL细胞在正常牙根面(N组)上的附着数量均明显多于病变(P组)根面(P<0.01);与对照(C组)相比,PDGF-BB(S组)既能促进PDL细胞在正常根面的附着(P<0.01),也能促进PDL细胞在牙周病变根面上的附着(P<0.05)。不加PDGF-BB时,PDL细胞在NC上的增殖率为80.0%,PC只有50%;加入PDGF-BB后,NS增殖率为249.4%,PS的增殖率为213.09%,与对照(C)组相比,PDGF-BB既能明显促进PDL细胞在正常根面的增殖(P<0.01),又能显著提高PDL细胞在病变牙根面上的增殖(P<0.01);扫描电镜下病变牙根面上生长的PDL细胞数量较少,形态细长,细胞的突起伸展不够充分,边缘皱缩,与根面附着较松散,经PDGF-BB作用后,细胞与根面的附着状态有所改善,较为紧密,数量有所增加。结论:PDGF-BB是一种有效的促PDL细胞生长的因子,可促进PDL细胞在牙根面的附着和增殖。
Objective:To evaluate the effects of platelet-derived growth factor-BB (PDGF-BB) on the attachment and proliferation of periodontal ligament ( PDL ) cells on different root surfaces. Methods: The attachment and proliferation of PDL cells on root surfaces of extracted teeth with or without periodontal disease was studied using cell culture and scanning electronic microscopy. Results: The PDL cells attached more on root surface of normal teeth ( Group N) than on affected teeth ( Group P) either with or without addition of PDGF-BB ( P 〈 0. 05 ). Compared with controls (Group C), PDGF-BB (Group S) stimulated the attachment of PDL cells both on normal root surface (P 〈 0. 01 ) and affected root surface (P 〈 0. 05 ). The rates of PDL cell proliferation were 80% and 50% respectively on groups NC and PC without PDGF-BB, and when PDGF-BB was added, rates of PDL cell proliferation in groups NS and PS were 249. 4% and 213. 09%. Compared with controls ( Group C ) , PDGF-BB significantly stimulated the proliferation of PDL cells both on normal root surface ( P 〈 0. 01 ) and affected root surface ( P 〈 0. 01 ). As shown by scanning electronic microscopy, PDL cells on affected root surface were less in quantity, slender in shape, with inadequate extension of cellular dendrites and shrinkage of cell borders. Stimulation with PDGF-BB markedly improved the cell attachment to the root surface and numbers of the cells. Conclusions: PDGF-BB appeared to be a positive growth factor to promote the attachment and proliferation of PDL cells.
