摘要
目的 用蛋白质组学相关技术分析难免流产与正常早孕子宫内膜蛋白质组的差异,探讨自然流产发生的可能机制。方法 用固相pH梯度双向凝胶电泳(2D—PAGE)分别分离难免流产(研究组)者和正常早孕行人工流产(对照组)者子宫内膜的蛋白质组。银染显色,PDQuest2DE软件分析。结果 图像分析测得2种胶的匹配率达84.5%,在等电点pI4~9、分子量17.0~75.4kD范围内分离得研究组子宫内膜蛋白点大约730个,对照组子宫内膜蛋白点大约780个,其中至少60个蛋白点在2种子宫内膜间有2倍以上的量变。结论 难免流产与正常早孕子宫内膜蛋白质组的差异表达,可能是导致孕卵着床胚泡植入障碍造成流产的一个原因。
Objective To examine the difference of proteomic expression of endometrium in inevitable abortion and normal early pregnancy so as to explore the potential mechanism of spontaneous abortion. Methods Proteoms of endometrium in inevitable abortion and normal early pregnancy were separated by immobilized PH gradient two-dimensional electrophoresis, (2-DE) and were analysed by PD Quest 2DE software after sliver staining. Results The matching rate of two gelatins is 84.5 %. At the isoelectric point of PH 4~9 and molecular weight of 17. 0~75. 4 kD, 730 protein spots were seperated in case group and 780 separated in control group. At least 60 protein spots had significantly differential expression between two groups. Conclusion The different proteomic expression of endometrium between inevitable abortion group and normal early pregnancy group may be involved in blastocyst implantation which leads to abortion.
