摘要
KAI1/CD82是近年来发现并分离出的一个新的肿瘤转移抑制基因,属于TM4SF(transmembrane4superfamily)家族成员,能与多种转录因子结合,调节基因转录。研究表明,KAI1/CD82在细胞膜上可与各种整合素分子、表皮生长因子受体组成复合体,通过PKC、IP4等信号通道来介导细胞的黏附,使肿瘤细胞不易脱离原发灶而起抑制转移功能,同时对肿瘤细胞的迁移和在转移部位的增殖有抑制作用,从而实现抑制肿瘤细胞转移和侵袭。本文就KAI1/CD82基因的结构、作用机制及抑制肿瘤转移及意义作一综述。
KAl1/CD82 gene was one of lately identified and isolated tumor metastasis inhibitor genes. It belongs to the transmembrane 4 superfamily (TM4SF) and interacts with many transcription factors (IF) to regulate gene transcription. Current data have shown that KAl1/CD82 could combine with many integrin members and epithelium growth factor receptors to form complex which mediate cell adherence via PKC, IP4 signaling, so it could prevent the detachment of tumor ceils from their primary focus; it could also inhibit tumor cells migration and proliferation in the secondary focus, and thus plays important roles in inhibiting the tumor metastasis and invasion. In this review, we focus on the gene structure of KAl1/CD82, its mechanisms as well as its future expectations in relation to tumor metastasis inhibition.
