摘要
目的研究硒(Se)和去甲肾上腺素(norep inephrine NE)对体外培养的心肌成纤维细胞(card iacfibrob lastCFbs)合成Ⅰ型胶原合成和mRNA表达的影响。方法采用酶消化法获得W istar大乳鼠CFbs并进行培养,与NE和Se(Selen ium)作用48 h后,测定细胞培养液中Ⅰ型胶原和羟脯氨酸含量,利用RT-PCR技术半定量分析细胞中mRNA表达情况。结果NE组上清液中的Ⅰ型胶原含量(A值)与对照组比较有显著性增加(P<0.01);补Se组A值较NE组显著性降低(P<0.01);补Se可降低NE引起的羟脯氨酸含量增加,且有显著性差异(P<0.01)。结论NE可以促进Ⅰ型胶原含量和CFbsα1Ⅰ型胶原mRNA表达增加,而Se对于Ⅰ型胶原的合成具有抑制作用。
Objective To investigate the effect of selenium(Se) on collagen Ⅰ synthesizing metabolism and mRNA expression in cultured Cardiac fibroblasts (CFbs) treated with Norepinephrine (NE). Methods CFbs were obtained by the method of enzymatic dispersion culture. Cell and culture medium were collected after 48 h, then examining the hydroxyproline (HYP) and collagen Ⅰ content as well as the expression of α1 [Ⅰ] collagen mRNA. Results The concentration of HYP in culture medium was obviously higher in NE group than that in the control( P 〈 0.01), however, the concentration of HYP in culture medium significantly reduced in cells treated with Se + NE compared with that in the NE group( P 〈 0.01 ) ; The level of collagen Ⅰ mRNA in culture medium was obviously higher in NE group than that in the control( P 〈 0.01), and the level of collagen Ⅰ mRNA in culture medium significantly reduced in cells treated with Se + NE compared with that in the NE group( P 〈0.01). Conclusions NE can increase the content of the HYP and Type Ⅰ collagen, however, Se could inhibit the synthesis and secretion of HYP and Type Ⅰ collagen, Se could also decrease the expression of α1 [Ⅰ] collagen mRNA.
