不同剂量活血祛瘀剂对小鼠免疫功能影响的差异 被引量:1

摘要 目的:分析活血祛瘀剂对小鼠细胞及体液免疫功能的影响。方法:实验于2005-01/06在泰山医学院分析实验室完成。选择昆明种小白鼠80只,按随机抽签法分为8组,即活血祛瘀剂3.0g/kg,1.5g/kg,0.75g/kg组、通塞脉片组、活血祛瘀剂3.0g/kg+环磷酰胺组、活血祛瘀剂1.5g/kg+环磷酰胺组、免疫低下模型组及正常对照组,每组10只。①活血祛瘀剂3.0g/kg,1.5g/kg,0.75g/kg组及活血祛瘀剂3.0g/kg+环磷酰胺组、活血祛瘀剂1.5g/kg+环磷酰胺组小鼠分别给予10mL/kg的活血祛瘀剂(主要有黄芪、丹参、葛根等组成)3.0g/kg,1.5g/kg,0.75g/kg,3.0g/kg,1.5g/kg灌胃,1次/d,连续14d。其中后两组小鼠均于给药后第3天腹腔注射环磷酰胺(75mg/kg)1次。②通塞脉片组给予1.05g/kg通塞脉片灌胃,1次/d,连续14d。③免疫低下模型组于实验第5天腹腔注射环磷酰胺(75mg/kg)1次,制备免疫低下模型。④正常对照组每日给予等量的生理盐水。通过巨噬细胞吞噬功能实验、溶血素水平(以吸光度413表示)测定及淋巴细胞转化率(以吸光度570~630表示)实验,观察活血祛瘀剂对小鼠免疫功能的影响。结果:纳入80只小鼠全部进入结果分析,无脱失。①各组小鼠巨噬细胞吞噬率比较:活血祛瘀剂3.0g/kg组显著高于正常对照组[(29.40±11.510)%,(19.10±9.231)%,(P<0.01)]。活血祛瘀剂3.0g/kg+环磷酰胺组、活血祛瘀剂1.5g/kg+环磷酰胺组均显著高于免疫低下模型组[(22.90±9.267)%,(19.50±6.485)%,(9.00±3.091)%,(P<0.01)]。②各组小鼠血清溶血素水平(吸光度413)比较:活血祛瘀剂3.0,0.75g/kg组均显著高于正常对照组[0.680±0.015,0.570±0.068,0.403±0.045,(P<0.01)]。活血祛瘀剂3.0g/kg+环磷酰胺组、活血祛瘀剂1.5g/kg+环磷酰胺组均显著高于免疫低下模型组[0.519±0.032,0.387±0.041,0.290±0.068,(P<0.01)]。③各组小鼠脾淋巴细胞转化率(吸光度570~630)比较:活血祛瘀剂3.0g/kg组显著高于正常对照组[0.530±0.060,0.458±0.048,(P<0.01)]。活血祛瘀剂3.0g/kg+环磷酰胺组,活血祛瘀剂1.5g/kg+环磷酰胺组均显著高于免疫低下模型组[0.647±0.036,0.555±0.038,0.225±0.043,(P<0.01)]。结论:不同剂量的活血祛瘀剂虽对正常小鼠免疫指标的影响有所不同,但对免疫抑制剂引起的细胞和体液免疫功能低下均有一定的保护和促进作用。 AIM: To explore the influence of huoxue quyu granule on cellular and humoral immunological function of mice. METHODS: The experiment was conducted at the analysis laboratory of Taishan Medical College between January and June 2005. Eighty Kunming white mice were selected and randomly divided into 8 groups: 3.0, 1.5 and 0.75 g/kg huoxue quyu granule groups; tongsaimai tablet group; high dose + cyclophosphamide group; middle dose + cyclophosphamide group; lowimmunity model group and control group with 10 mice in each group.① The mice in the 3.0, 1.5 and 0.75 g/kg huoxue quyu granule groups and high, middle dose+cyclophosphamide groups were intragastrically infused with 10 mL/kg huoxue quyu granules (mainly composed of huangzhi, dan- . shen, gegen and so on) 3.0 g/kg, 1.5 g/kg, 0.75g/kg, 3.0 g/kg, 1.5 g/kg, respectively, once a day for 14 days. Those in the latter two groups were intraperitoneally injected with cyclophosphamide (75 mg/kg) once on the 3^nd day after administration. ②Tongsaimai tablet group was intragastfically infused with 1.05 g/kg tongsaimai tablet, once daily for 14 days. ③The low-immunity model group was intraperitoneally injected with cyclophosphamide (75 mg/kg) to prepare the model of low-immunity.④The control group was given saline at the same dose every day. The influence of huoxue quyu granules on immunological function of mice was observed by macrophage phagocytic function test, quantitative hemolysis spectrophotometry (represented by A413) and lymphocyte transformation test (A570-630). RESULTS: Eighty mice were all involved in the result analysis, without dropout.①Comparison of phagocytosis rate of histocytes in mice of every group: The 3.0 g/kg huoxue quyu granule group was obviously higher than the control group [(29.40±11.510)%, (19.10±9.231)%, P 〈 0.01]. The high and low doses + cyclophosphamide groups were higher than the low-immunity model group [(22.90±9.267)%, (19.50±t.485)%, (9.00±3.091)%, P 〈 0.01].②Comparison of A413 in every group: The &13 of mice in the 3.0 g/kg and 0.75 g/kg granules groups was markedly higher than that in the control group (0.680±0.015, 0.570±0.068, 0.403±0.045, P 〈 0.01). The A413 in the high and low doses + cyclophosphamide groups were higher than the low-immunity model group (0.519±0.032, 0,387±0.041, 0.290±0.068, P 〈 0.01). ③Comparison of transformation efficiency of A(570-630). The A(570-630) in mice of the 3.0 g/kg granules group was obviously higher than the control group (0.530±0.060, 0.458±0.048, P 〈 0.01) and the high and low doses + cyclophosphamide groups were higher than the low-immunity model group close to that in the blank control group (0.647±0.036, 0.55±0.038, 0.225 ±0.043, P 〈 0.01). CONCLUSION: Although different doses of huoxue quyu granules can influence differently the immune indexes of normal mice, it has protective or immunoproliferative effect on cellular and humoral immunity caused by immunosuppressive agent.
