摘要
目的:鉴定复方五仁醇胶囊入血成分并进行含量测定,为该制剂血清药理学研究结果验证积累数据。方法:灌胃给药制备含药血清,高效液相法测定含药血清的色谱图,与体外制剂及对照品色谱图进行比较,分析、鉴定入血成分,并对鉴定出的主要成分进行含量测定。结果:灌胃给药后血清中产生了13个与药物因素有关的成分,其中8个为制剂原形成分,包括五味子醇甲、五味子甲素和五味子乙素,其余5个为制剂成分的代谢产物。血清中五味子醇甲、五味子乙素的平均含量分别为11.0632,7.4909mg·L-1。结论:五味子醇甲、五味子乙素在血清中含量较高,可能为复方五仁醇胶囊重要的药效成分。
OBJECTIVE To identify the constituents migrating to the blood of rats after oral administration of compound Wurenchun capsules and determine the content of these constituents, thus accumulate data for the validation of serum pharmacology of this preparation. METHODS Serum containing drug were prepared after the preparation oral administered to rats and the chromatogram of this serum was determined by HPLC. By comparing the chromatogram of serum containing drug with the ones of preparation and control compounds, constituents migrating to blood were identified and the main ones were determined. RESULTS Thirteen drug-induced constituents in blood were detected, eight of them were the ones contained in the preparation, including schisandrin, deoxyschizandrin and schisandrin B, five of them were metabolites. The average content of schisandrin, and schisandrin B in blood was 11.0632,7.4909mg·L^-1 respectively. CONCLUSION Schisandrin and schisan drin B account for higher than other in serum containing drug and they likely be the effective substances in compound Wurenchun capsules.
