已酮可可碱对SIRS大鼠核因子-κB活性的影响

摘要 目的探讨已酮可可碱(Pentoxifylline,PTX)对全身炎症反应综合征(Systemic inflammatory response syndrome,SIRS)大鼠核因子-κB(NF-κB)活性的影响。方法用内毒素复制SIRS大鼠。实验分三组,即正常对照组、SIRS组和PTX组。用ELISA的方法检测各组NF-κB活性和IL-6的表达。结果与正常对照组比较,SIRS组大鼠NF-κB活性和IL-6表达明显升高(P<0.01);与SIRS组比较,PTX组大鼠NF-κB活性和IL-6表达明显降低(P<0.01)。在SIRS组大鼠中NF-κB活性和IL-6表达呈明显正相关(P<0.01)。结论PTX可明显抑制SIRS大鼠的炎症反应,其作用机制可能是抑制NF-κB的活化。 Objective To explore the effects of pentoxifylline (PTX) on the activation of nuclear factor Kappa B ( NF - κB) of rats with systemic inflammatory response syndrome. Methods The animal model of SIRS was established by endotoxin. 60 Wistar rats were distributed into three groups randomly. They are the control group, SIRS group and PTX group, the activation of NF- κB and the expression of IL- 6 were examined by ELISA. Results Compared with the control group, the activation of NF - κB and the expression of IL - 6 in the SIRS group increased significantly ( P 〈 0. 01 ). Compared with the SIRS group, the activation of NF - κB and the expressions of IL- 6 in the PTX group decreased significantly (P 〈 0.01 ). In the SIRS group, there were significant positive correlation between the activation of NF - κB and the expression of IL - 6 ( P 〈 0.01 ). Conclusion PTX may alleviate the inflammatory reaction of SIRS by inhibiting NF - κB activation.
