球面对称设计法研究纤维素酶对枸杞子中枸杞总糖提取率的影响 被引量:4

摘要 目的研究纤维素酶对枸杞子中枸杞总糖提取率的影响,综合考虑水浴温度、水浴时间和酶加入量三方面因素,按照球面对称设计的要求确定最佳的提取条件。方法将纤维素酶液加入枸杞子匀浆中,用3,5-二硝基水杨酸(DNS)比色法测定枸杞子中总糖的含量。结果纤维素酶在水浴温度为60℃、水浴时间为300s、加入的酶液体积为5mL时可使枸杞子中枸杞总糖的提取量提高1.66%。结论纤维素酶可提高枸杞子中枸杞总糖提取率。 Objective To study the effect of cellulase on the extraction rate of medlar total saccharide, considering three factors of temperature, time and quantity of cellulose. The best extracting condition was determined according to the require of sphere symmetry design, Methods Add cellulase liquid into the medlar slurry and determine the content of medlar total saccharide by DNS method. Result The extraction rate of medlar total saccharide was increased 1.66% by cellulase at temperature of 60℃, time of 300 s and cellulase volume of 5 mL. Conclusion Cellulase can promote the extraction rate of medlar total saccharide.
