摘要 目的探讨基质金属蛋白酶-2(MMP-2)在脱氧皮质酮-盐型高血压大鼠(DHR)心脏微小血管重塑中的作用及麝香保心丸对其的影响。方法18只雄性SD大鼠切除左侧肾脏后1周,随机等分为三组。建立DOCA-HDR模型,5周后检测心脏血管密度以及MMP-2因子的表达量。结果在DHR左心室心内膜下心肌中存在微小动脉密度增加和毛细血管密度减少,MMP-2mRNA和MMP-2/TIMP-2的蛋白表达上调;麝香保心丸不能抑制血压升高,但能减轻微小血管损害,下调MMP-2mRNA和蛋白表达,MMP-2的表达同微小血管密度间具有良好的相关性。结论DHR心脏中存在微小血管病变,MMP-2/TIMP-2表达可能参与微小血管病变的病理过程,麝香保心丸可能通过调节MMP-2表达产生对DOCA-高血压大鼠心脏的微小血管损害的保护作用。 Ojective:To investigate the cardiac microvaular density, the expression of MMP- 2 mRNA and protein, and the effects of Shexiangbaoxin Pill(麝 香 保 心 丸 ) on them in DOCA- salt Hypertensive Rats.Methods:18 male Sprague- Dawley rats were unilaterally neprectomiled,and were divided into 3 groups including control, placebo,and Shexiangbaoxin Pill group. They were plot to death after 5 weeks treatment. The arteriolar and capiullary density, and the expression of MMP- 2/TIMP- 2 protein were employed to evaluate in the subendocardial myocardium of the left ventricular tissue. RT- PCR was used to measure the expression of mRNA of MMP- 2/TIMP- 2.Results:DOCA- Salt hypertensive rats exhibited a marked change of microvascular density, and an increase of expression of MMP- 2/TIMP- 2.Shexiangbaoxin Pill had no effect on arterial pressure, but decreased microvasular density and downregulated the expression of MMP- 2/TIMP- 2.Conclusions:MMP- 2 may play an important role in cardiac microvascular density of DOCA Salt Hypertensive Rats, Shexiangbaoxin Pill may inhibit cardiac microvascular change by regulating the expression of MMP- 2/TIMP- 2.
