摘要
目的观察CpG-ODN 2006和CpG-ODN 2216对健康人外周血单个核细胞中B淋巴细胞抗原递呈相关分子MHC-I、MHC-Ⅱ及共刺激分子CD 80、CD 86表达的影响,探讨CpG-ODN对B淋巴细胞抗原递呈功能的作用。方法CpG-ODN 2006、CpG-ODN 2216与健康人PBM C共培养48 h,用流式细胞技术以CD 19-P reCP“设门”其中B淋巴细胞,检测其表面CD 69、MHC-Ⅰ、MHC-Ⅱ、CD 80及CD 86的表达状况,并与未加CpG-ODN组进行比较。B淋巴细胞MHC-I、MHC-Ⅱ的表达量以几何平均荧光强度表示(M F I);CD 80、CD 86的表达量以阳性细胞百分率表示。结果CpD-ODN 2006和CpG-ODN 2216均显著提高B淋巴细胞CD 69、MHC-Ⅰ、MHC-Ⅱ、CD 80及CD 86的表达,与未加CpG-ODN组比较有显著性差异(P<0.05)。CpG-ODN 2216使B淋巴细胞表面抗原递呈相关分子的增加量更为明显。结论CpG-ODN能够提高B淋巴细胞表面抗原递呈相关分子和共刺激分子的表达,能增强B淋巴细胞递呈抗原的能力。
Objective To investigate the potential of immunostimulatory CpG-ODN2006 and CpG-ODN2216 to enhance surface molecules expression on human peripheral blood B lymphocyte,such as MHC-Ⅰ ,MHC-Ⅱ ,CD80 and CD86,which is associated with antigen processing. Methods The PBMCs were incubated with CpG-ODN2006 or CpG-ODN2216 after a 48-hour period,the expression of surface molecules CD69,MHC-Ⅰ ,MHC- Ⅱ ,CD80 and CD86 on B lymphocyte were examined by gatting CD19 positive cells by FACS analysis. The results were compared with control group. The expression of MHC-Ⅰ and MHC- Ⅱ on B lymphocyte was described as the mean fluorescence intensity (MFI),and the expression of CD80 and CD86 on B lymphocyte was described as the percents in all B lymphocyte. Results Expression of surface molecules CD69,MHC-Ⅰ ,MHC-Ⅱ ,CD80 and CD86 on B lymphocyte was up-regulated observably by CpG-ODN2006 or by CpG-ODN2216. There was significantly difference between the group of CpG-ODN and the group of control (P^0. 05). Conclusion CpG-ODN trigger a strong up-regulation of surface molecules associated with antigen processing on human B lymphocyte. All these surface molecules are major importance for professional APC function.
