摘要 目的:观察骨髓间充质干细胞移植对心肌梗死后心肌细胞凋亡的影响,探讨干细胞移植改善心功能机制。方法:实验于2004-09/2005-10在郧阳医学院附属十堰市太和医院生命科学研究所完成。①21只日本大耳白兔采用随机数字法分为正常对照组、心肌梗死组及干细胞移植组,每组7只。②心肌梗死模型制作3周后,心肌梗死组及干细胞移植组分别在梗死周边区域注射100μL细胞培养基或干细胞悬液,对照组不处理。③4周后心脏超声检测心功能、左室收缩末期内径、左室舒张末期内径等评价心室重塑。处死动物后经流式细胞仪检查梗死区域细胞凋亡情况。结果:①各组兔心脏超声检测结果:与心肌梗死组相比,干细胞移植治疗后,左心室射血分数[(56.27±1.73),(69.41±3.19)%,P<0.001]、左室收缩末期内径[(10.25±0.89),(8.13±0.79)mm,P<0.001]、左室舒张末期内径[(13.82±1.02),(11.24±1.21)mm,P<0.001]、室壁运动明显改善。②各组细胞凋亡检查结果:干细胞移植后,细胞凋亡显著降低[(14.00±0.31)%,(4.30±0.57)%,P<0.001]。结论:干细胞移植可抑制细胞凋亡,改善心功能,预防心室重塑,具有改善梗死后心脏功能的作用。 AIM: To observe the effect of mesenchymal stem cells (MSCs) on the myocardial cell apoptosis after myocardial infarction, and explore the improved cardiac function of stem cells. METHODS: The experiment was performed at the Institute of Life Science, Shiyan Taihe Hospital Affiliated to Yunyang Medical College from September 2004 to October 2005. ①A total of 21 Japanese flap-eared rabbits were divided into normal control group, myocardial infarction group and stem cell group with 7 rats in each group.②100 μL cell medium or stem cell suspension was implanted in the myocardial infarction group and stem cell group 3 weeks after the model of myocardial infarction were made. Control group did not receive any intervention, ③After 4 weeks, cardiac function, bore in left ventricular end-systolic stage and bore in left ventricular end-diastolic stage were measured with heart ultrasound to assess the ventricular remodeling. Apoptosis in infarcted region was examined with flow cytometer after killing animals. RESULTS: ① Result of cardiac ultrasound in rabbits of each group: Compared with myocardial infarction group, left ventricular ejection fraction [(56.27±1.73), (69.41±3.19)%, P 〈 0.001], the bore of left ventricular end-systolic stage [(10.25±0.89), (8.13±0.79)mm, P 〈 0.001], bore in left ventricular end-diastolic stage [(13.82±1.02), (11.24±1.21) mm,P 〈 0.001] and ventricular wall motion were markedly improved after stem cell transplantation. ②Result of apoptosis examination: After stem cell transplantation, the apoptosis decreased obviously [(14.00±0.31)%, (4.30±0.57)%,P 〈 0.001]. CONCLUSION: Stem cell transplantation can inhibit cell apoptosis, improve cardiac function, provent ventricular remodeling and ameliorate cardiac function after infarction.
