摘要 目的:探讨增殖细胞核抗原(PCNA)在大鼠肾上腺细胞培养过程中的表达及其意义。方法:应用SP免疫组化法,检测PCNA在大鼠肾上腺细胞培养不同时期表达的强度变化,同时测定肾上腺细胞分泌皮质酮功能的动态变化。结果:在肾上腺细胞体外培养过程中,第3、5、7天,PCNA蛋白表达逐渐增强,分别为(18±3)%、(38±5)%、(70±6)%,第9、11天与第7天相比,PCNA蛋白表达减弱(P<0.05),皮质酮浓度测定显示在第7天皮质酮浓度达到最高峰(101.2±15.8nmol/L),随后逐渐下降,与PCNA蛋白表达强度的变化趋势一致。结论:肾上腺细胞体外培养在第7天肾上腺细胞增殖最旺盛,而且肾上腺分泌皮质酮达到最高峰。 Objective: To explore expression and significance of PCNA in rats adrenocortical cell culture in vitro. Metbods:Immunohistochemical methods were adopted, expressing strengths of PCNA were examined in ceil culture in different phase, meanwhile, corticosterone concentrations were dynamically measured. Results: In adrenocortical cell culture in vitro expressions of PCNA were increasing in the third, fifth or seventh day( 18±3 %, 38 ±5%, 70±6% respectively), in contrast to the ninth or eleven day PCNA gradually decreased(50±3%、39±2%, P 〈0.05). corticosterone measurements showed corticosterone concentration was peak in the seventh day (101.2±15.8 nmol/L), and gradually degraded. Changes of PCNA expressing strengths were consistent with those of corticosterone concentrations. Conclusions: Adrenocortical cells were the most proliferative in vitro and excreting corticosterone was at the most.
