小剂量吲达帕胺/阿替洛尔联用对自发性高血压大鼠降压和改善心功能的作用

摘要 目的比较不同剂量吲达帕胺(Ind)和阿替洛尔(Ate)联用对自发性高血压大鼠(SHR)的降压作用及对心功能的影响。方法48只SHR随机分为6组。①空白对照组;②低-低剂量组:Ind0.06mg/kg·d-1,Ate2.25mg/kg·d-1;③低-高剂量组:Ind0.06mg/kg·d-1,Ate4.5mg/kg·d-1;④高-低剂量组:Ind0.12mg/kg·d-1,Ate4.5mg/kg·d-1;⑤高-高剂量组:Ind0.12mg/kg·d-1,Ate4.5mg/kg·d-1;⑥高剂量单用组:Ate4.5mg/kg·d-1。灌胃给药,连续给药5周,尾袖法测收缩压,Langendorff灌流心脏测量心功能。结果高-低剂量联用组,有效降低SHR的收缩压,降低左室舒张末期压,减少左室重与体重之比。结论小剂量联合应用Ind和Ate对SHR的降压作用及心功能的改善优于大剂量单用Ate。 Objective The purpose of this research is to study the effect of SBP reduction and cardiac function after treatment with different doses of combined indapamlde and atenolol in spontaneously hypertensive rat (SHR). Method 48 SHR were randomly divided into 6 groups. ① Untreated group.② Low-low dose combination: Ind 0.06 mg/kg and Ate 2.25 mg/kg per day.③ Low-high dose combination: Ind 0.06 mg/kg and Ate 4.5 mg/kg per day.④ High-low dose combination: Ind 0.12 mg/kg and Ate 2.25 mg/kg per day.⑤High-High dose combination: Ind 0.12 mg/kg and Ate 4.5 mg/kg per day.⑥ High dose Ate monotherapy: Ate 7.14 mg/kg per day. All SHR were treated with drugs by gavage method for successive 5 weeks. The tail arterial SBP was measured once per week. Cardiac function was assessed by isolated Langendorff heart perfusion system. Result All 5 treatment groups showed a significant decrease in SBP after the therapy (P 〈 0.01 ). The ratio of left ventricular weight to body weight in Ind-H/Ate-L group was significantly decreased by 12% when compared with the Ate monotherapy group (P〈 0.05). Conclusion Combination of a low dose of indapamlde and atenolol as well as a high dose atenolol monotherapy significantly decreased the SBP of SHR and inhibited LVH development.
