小鼠脾细胞CRF受体放射配体测定

Radioligand assay on CRF receptor of mouse splenocytes

作者采用放射配体分析法，检测了小鼠脾细胞ＣＲＦ特异性结合受体。实验结果表明，受休测定的较适条件为２５Ｃ，ｐＨ７．４，反应时间２０ｍｉｎ，脾细胞数为４×１０~６个／管。^(１２５)Ｉ一ＣＲＦ结合脾细胞的Ｓｃａｔｃｈａｒｄ作图分析显示，脾细胞上存在有高亲和力ＣＲＦ受体，其ｋｄ值为１．６ｎｍｏｌ／Ｌ。脾细胞上神经肽ＣＲＦ受体的检出。

A radioligand assay for CRF receptor of mouse splenocytes was developed using125I-Tyr-CRF as the ligand.The right condition was incubation at 25℃,pH 7.4,for 20min in 4 × l06splenocytes/tub. By scatchard plot,it was shown that there was a high-affinity binding sites to CRFfor kd of l.6 nmol/L on mouse splenocytes. These findings suggest that neuropeptide CRF could in-fluence on immunocyte function.
