氧自由基对胃癌细胞MGC-803的P53、P21ras蛋白表达的影响

摘要 目的:观察氧自由基对人胃癌细胞系MGC-803细胞的P53、P21ras蛋白表达的影响,探讨氧自由基在胃癌发生发展中的作用。方法:通过细胞培养、采用硫代巴比妥酸反应物质法检测细胞内氧自由基中间产物丙二醛(MDA)的含量,应用免疫组织化学技术检测MGC-803细胞内不同浓度的氧自由基对P53、P21ras蛋白表达的影响。结果:氧自由基含量升高可增强MGC-803细胞内P53、P21ras蛋白表达(P<0.01),氧自由基水平降低则P53、P21ras蛋白表达减弱(P<0.01)。结论:胃癌细胞系MGC-803内氧自由基水平的改变可影响其P53、P21ras蛋白表达。 Objective: To study the effect of oxygen free radical (OFR)on P53 and P21ras protein expressions in human sastric cancer cell line MGC-803 and the role of OFR in development of gastric cancer. Methods: The content of malondiatdehyde (MDA) in cuhured human gastric cancer line MGC-803 cells was measured as intermediate product of OFR by thiobarbiturie acid reactive substances(TBARS) method. Immunohistochemical staining (IHC) was used to detect the expressions of P53 and P21ras proteins in the MGC-803 cells treated with different levels of OFR. Results: The expressions of P53 and P21ras proteins promoted when OFR level was raised in the MGC-803 cells (P 〈 0.01), and those decreased when OFR level decreased (P〈0.01). Conclusion: Different levels of OFR can influence the expressions of P53 and P21ras proteins in gastric cancer cell line MGC-803.
