Expression of Marrow Vascular Endothelial Growth Factor and Proliferating Cell Nuclear Antigen in Children with Acute Leukemia and Their Clinical Significance

摘要 目的探讨增殖细胞核抗原(PCNA)和血管内皮生长因子(VEGF)在儿童急性白血病(AL)中的表达及其意义。方法采用S-P免疫组织化学方法,检测59例AL患儿骨髓细胞PCNA和VEGF表达,比较不同类型白血病、初治组与复发难治组、治疗缓解组与未缓解组间的表达差别。选择同期住院15例非白血病患者正常骨髓作为对照组。结果PCNA、VEGF在AL患儿中表达均明显高于对照组(t=5.59,6.67 P均<0.05);PCNA在初治ALL中表达显著高于ANLL(t=2.681 P<0.05),VEGF表达无差异(t=0.970 P>0.05);初治组与复发难治组、缓解组与未缓解组两者表达均有显著性差异(P均<0.05)。结论PCNA和VEGF在儿童AL中存在高表达,以不同机制参与AL发生、发展,与疾病预后有关。 Objective To explore the expression of vascular endothelial growth factor(VEGF) and proliferating cell nuclear antigen (PCNA) in children with acute leukemia(AL) and their clinical significance. Methods The expression of VEGF and PCNA in bone marrow cell of 59 cases of AL were detected by S-P immtmohistochemical staining, their expression difference were compared between ALL and ANLL, the initial treatment group and the recurrence and refractory group, remission group and no-remission group. In the same period, 15 cases of normal bone marrow cell were selected as control group with no-leukemia. Results The expression of VEGF and PCNA in AL were significantly higher than those in the control group(t= 5.59,6.67 all P〈0.05); In the initial treatment patients, PCNA of ALL group and ANLL group showed significant difference(t=2.681 P〈0.05), while VEGF of ALL group and ANLL group showed no significant difference( t=0.970 P〉0.05) ; In both PCNA and VEGF, there were significant difference between initial treatment group and recurrent and refractory group, remission group and no-remission group(all P〈0.05). Conclusions VEGF and PCNA expression in children with AL are high, and having different roles in their occurrence and development of AL. It may be helpful to evaluate prognosis of AL by detecting them jointly.
