c-fos反义寡聚核苷酸对癫癎发作后细胞凋亡的影响 被引量:4

Effect of c-fos Antisense on Neuronal Apoptosis in Rats′ Brain with Pentyleneterazol-Induced Seizure

摘要 目的探讨c-fos基因在戊四氮(PTZ)致大鼠模型中表达及其与细胞凋亡的关系。方法采用免疫组织化学法检测致模型大鼠海马神经细胞Fos表达;TUNEL及流式细胞仪检测细胞凋亡;c-fos反义寡聚核苷酸侧脑室注射干预,观察其在抗凋亡中作用。结果PTZ致后大鼠可引起Fos在脑内一过性高表达,1 h达高峰(P<0.001),而神经细胞凋亡在24 h达峰值(P<0.001);c-fos反义寡聚核苷酸部分抑制Fos表达,并显著减少神经细胞凋亡。结论癫发作后Fos早期表达可能与后期神经细胞凋亡有关;c-fos反义寡聚核苷酸能抑制脑内Fos表达,对癫造成脑损伤起一定保护作用。 Objective To probe the effect of c-fos gene on the pentyleneterazol-induced hippocampal neurons apoptosis. Methods Using immunohistochemistry,TUNEL and flow cytometry(FOM), we detected the Fos exprexsion and the apoptosis of hippocampal neurons;we injected c-fos antisense into ventrile before epilepsy and detected as up. Results Epilepsy can induce the expression of Fcxs in the hippocampus and peaking at 1 h( P〈0.001 ) ; TUNEL and FCM show that the apoptotic neurons began to increase at 12 h, peaking at 24 h( P〈0.001 ) ; Microinjecting c-fos antisense into ventricle 15 h before PTZ the expression of Fos was nearly disappeared and the apoptosis also decreased, but NS has no this kind of influence. Conclusions The expression of c-fos may play an important role in apoptosis of epilepsy by itself or other product who induced; c-fos antisense can decrease the apoptosis by decreasing the expression of c-fos, and may become a hoping method in treating epilepsy in the future.
