摘要
目的 建立测定氢定乳膏中氢化可的松和醋酸氯己定含量的方法。方法 采用高效液相色谱法。分析 色谱柱为Extend C18(4.6mm×150mm,5μm),流动相为甲醇-水相(醋酸铵26g和三乙胺37mL,加水至1000mL,冰醋酸调pH至4.0)(55:45)为流动相;流速为1.0mL·min^-1;检测波长为254nm;柱温为30℃;进样量为10止。结果 该方法不受处方中基质干扰,氢化可的松在0.1~0.6mg·L^-1内和醋酸氯己定在0.02~0.12mg·L^-1内,峰面积与质量浓度呈良好的线性关系(r hydrocortisone=0.9998,r chlohexidine acetate=0.9998),平均回收率:氢化可的松为99.6%、醋酸氯己定为99.2%。RSD分别为0.34%,1.41%(n=9)。结论 该方法可用于氢定乳膏中氢化可的松和醋酸氯己定的含量测定。
OBJECTIVE To develop a method for the determination of hydrocortisone and chlohexidine acetate in Qingding cream. METHODS A high-performance liquid chromatography method was used with Extend C18 column.The mobile phase consisted of methanolaqueous phase (ammonium acetate 26g and triethylamine 37 mL,imbitition to 1 000 mL, adjust pH to 4.0 by glacial acetic acid) (55:45) and the flow rate was 1.0 mL·min^-1, with the detection wavelength at 254nm.RESULTS The matrix component in cream didn't interfere with determination of hydrocortisone and chlohexidine acetate. The calibration curve was linear within the range of 0.1 - 0.6mg·L^-1 for hydrocortisone( r = 0.999 8) and the range of 0.02 - 0.12mg·L^-1 for chlohexidine acetate( r = 0.999 8) .The average recoveries of hydrocortisone and chlohexidine acetate were 99.6% and 99.2% respectively. RSD were 0.34% and 1.41% ( n = 9), respectively. CONCLUSION The method can be used for the determination of hydrocortisone and chlohexidine acetate in Qingding Cream.
