摘要
使用Wistar大鼠,暴露左侧MCA并分别夹闭3、6、12、24h,再灌2h;另一部分夹闭6h,分别再灌4、8、12、16h。结果表明,左侧脑电图明显受抑,TTC染色显示缺血,再灌的脑皮质损伤区不着色,并随缺血和再灌时间延长而扩大。光镜提示,随缺血时间延长,脑组织充血,水肿加重,神经细胞变性,出血,坏死及海马损伤。缺血6h随再灌时间延长脑组织有灶性出血和软化灶形成,并有神经细胞核固缩,少许核破裂。证明阻断MCA中段接近嗅束外侧所建立的局灶性脑缺血-再灌损伤的动物模型是成功的,值得在实验研究中应用。
A small hole of 2~3mm in diameter was drilled on the left sphenoid bone of Wistar rat.In 1 group,left middlecerebral artery(MCA)was exposed and clipped for 3,6,12,24 hours,and followed by 2 hours of reperfusion.In the sec-ond group MCA was clipped for 6 hours and followed by 4,8,12,16 hours of reperfusion.The electroncephalographshowed inhibition in activity of the left brain.After perfusing the animal with gelatin-carbonizing ink the clipped left brainwere colorless.Whereas opposite side was staining black.TTC staining of the brain sectionals showed that the injured re-gion of cortex was uncoloured,as the time of ischemia and reperfusion prolonged.In histological examination cogestion andedama in brain tissue and hippocampus damage were getting worse,as the ischemia prolonged.If the ischemia occurred formore than 6 hours focal bleeding and softening of the brain tissue appearid together with karyopyknosis and a few karyor-rhexis.The report indicates that animal the model of focal brain ischemiareperfusion was successfully established with clip-ping of the MCA at the outer olfactory sulcus.The operative precedure is relatively simple.It is useful in studying focalbrain ischemiareperfusion injury.
