L-(一)-苹果酸经酯化、还原、羟基保护、缩合、羰基还原、羟基保护、脱硅保护和Swern氧化等反应制得罗伐他汀的关键中间体(3R,5S)-6-氧代-3,5-O-亚异丙基-3,5-二羟基己酸叔丁酯,总收率25%。
t-Butyl (3R,5S) -6-oxo-3,5-O-isopropylidene-3,5-dihydroxyhexanoate, a key intermediate of rosuvastatin, was synthesized from L- (-) malic acid by esterification, reduction, hydroxyl protection, condensation, carbonyl group reduction, hydroxyl protection, deprotection and Swern oxidation with an overall yield of 25%.
