Epstein-Barr病毒潜伏膜蛋白基因免疫的实验研究

摘要 利用基因免疫技术,将重组质粒pBs-LMP-Hyg注入BALB/C小鼠骨骼肌中,于第2、4、8周用间接免疫荧光法检测鼠血清中EB病毒潜伏膜蛋白(LMP)特异抗体。结果:所有免疫小鼠(5/5)均产生特异抗体,平均抗体滴度第2周为1/16,第4周为1/32,第8周为1/44.8,质粒注射8周后的肌细胞中仍可扩增出目的基因DNA序列。提示:目的基因至少可在受体组织中存在并表达到第8周,质粒DNA1次性免疫后抗体滴度随时间变化逐渐增高。 For the purpose of gene immunization, we injected the expression plasmid pBS-LMP-Hyg directly into the quadriceps muscle of BALB/C mice. Indirect immuno-fluorescence assay was used to detect specific antibody against EBV latent membrane protein(LMP) in sera of mice, at the second, fourth and eighth weeks, The results showed that all the mice were able to elicit specific immune responses and the antibody titres in mice rose along with the time, Plasmid DNA existed and was expressed in receptive tissue at least for eight weeks.
