支原体确证琼脂培养基检测泌尿生殖道支原体的优势分析

Advantages of Application of Mycoplasma Verifying Agar in Detection of Genital Mycoplasmas

目的评价支原体确证琼脂培养基在诊断泌尿生殖道标本中解脲支原体(Ureaplasmaurealyticum,Uu)和人型支原体(Mycoplsmahominis,Mh)的优势,为提高临床泌尿生殖道支原体感染实验室诊断的准确性提供依据。方法取经过12小时UU/MH肉汤增菌的临床疑似标本洗脱液直接接种于支原体确证琼脂培养基,在5%CO2和95%N2的气体环境中培养24～48h后,低倍镜下观察特征性菌落,计数其菌落形成单位(ColonyFormingUnits,CFU)。同时用支原体药敏试剂盒作对照,观察UU孔、Mh孔及计数孔颜色的变化。结果172例泌尿生殖道标本中,支原体确证琼脂培养基检出UU感染59例,MH感染19例,混合感染16例,UU计数≥104CFU者40例,MH计数≥104CFU者15例;支原体药敏试剂盒检出UU感染63例,MH感染13例,混合感染13例,UU计数≥104CFU者45例,MH计数≥104CFU者10例。结论支原体确证琼脂培养基,简便易行,观察菌落准确性高,提高MH的检出率及液体培养基的确证性。

Objective To assess the advantages of Mycoplasma Verifying Agar（MVA） in diagnosis of Ureaplasma urealyticum（UU） and Mycoplasma homminis（MH） from infected urogenital tract, so as to improve the accuracy of clinical Mycoplasma laboratory diagnosis by providing experiment data. Methods Rinse the swab in the UU/MH broth and keep the vial at 36. 5℃ for 12 hrs, and then inoculate the broth into MVA, and incubate at 5% CO2/95%N2 for 24 to 48 hrs. Observe the typical colonies with lowly amplified microscope and count the colony forming units（CFU）.Do the control experiment with Mycoplasma Susceptibilities Test Kit（MSTK） by observing the color changes. Results Of all 172 cases of urogenital samples, MVA shows 59 cases of UU, 19 cases of MH, 16 cases of UU and MH, 40 cases of UU count≥10^4CFU, and 15 cases of MH count≥10^4CFU, by contrast, MSTK indicates 63 cases of UU, 13 cases of MH, 13 cases of UU and MH, 45 cases of UU count≥10^4CFU, and 10 cases of MH count≥10^4CFU. Conclusions MVA is a simple and easy diagnostic method with high accuracy by directly observing specific colonies It can improve the accuracy of liquid culture media and do better than liquid culture media in detection of Mycoplasma hominis.
