摘要
目的:观察层粘连蛋白总糖肽(Lamininglycopeptides,LN-Gps)对小鼠腹腔巨噬细胞(M)杀伤黑色素瘤B16细胞及吞噬抗体包被的羊红细胞(EA)作用的影响。方法:将BALB/c小鼠腹腔M体外培养48h后,再分别与0(对照),50,100,200μg/ml终浓度的LN-Gps预温30min,用噻唑蓝比色法检测M对B16细胞的杀伤作用,用联苯胺氧化比色法检测M吞噬EA的功能。结果:(1)M杀伤B16细胞的作用呈剂量依赖性增高,其中100μg/ml和200μg/ml的LN-Gps可分别使M对B16细胞的杀伤作用较对照组增高(10.3±2.8)%和(28.4±9.9)%(P均<0.05)。(2)M对EA吞噬作用降低,分别为对照组的(92.2±12.4)%(LN-Gps50μg/ml)、(87.0±14.2)%(LN-Gps100μg/ml,P<0.05)和(69.4±18.2)%(LN-Gps200μg/ml,P<0.05)。(3)如果将B16细胞与LN-Gps200μg/ml先预温,则M对B16细胞的杀伤作用较对照组(B16细胞不与LN-Gps预温)明显下降,为对照组的(84.7±7.3)%(P?
Objective:To explore the effects of laminin glycopep des on the macrophage functions of cytotoxicity against B16 cells and phagocytosis of opsonized red cells.Methods:The macrophages,collected from BALB/c mouse peritoneal lumen, were cultured in vitro.After 48 h,LN-Gps were added at the final concentrations of 50, 100, and 200 μg/ml,37℃,30 min later,the M cytotoxicity to B16 target cell was measured by MTT colorimetric assay,and the M phagocytosis of EA was recorded using a microplate spectrophotometer.Results:(1) The macrophage cytotoxicity against B16 cells was enhanced as the levels of LN-Gps were elevated.When the macrophages were pre-exposed to 100 μg/ml and 200 μg/ml LN-Gps,their cytotoxicity against B16 cells were elevated by(10.3±2.8)%(P<0.05) and(28.4±9.9)%(P<0.05),respectively,as compared with the control.(2) The phagocytosis of opsonized red cells by macrophages decreased after the macrophages were pre-exposed to LN-Gps,the phagocytosis of treated groups were(92.2±12.4)% (LN-Gps 50 μg/ml),(87.0±14.2)% (LN-Gps 100 μg/ml,P<0.05) and(69.4±18.2)% (LN-Gps 200 μg/ml,P<0.05) of the control group.(3) If the B16 cells were pre-treated,while the macrophages were not treated with 2 0 0 μg/ml LN-Gps,the cytotoxicity of macrophage against B16 cells was significantly inhibited,only(84.7±7.3)% of the control group(P<0.05).Conclusion:The effects of laminin glycopeptides were not parallel in vitro.As to different cells,or to different functions of certain cells,the LN-Gps have different effects.The mechanisms are worth further studying.
