42例脑星形胶质细胞瘤P53蛋白表达及与PCNA的关系 被引量:1

摘要 目的:观察人脑星形胶质细胞瘤中P53蛋白的表达及与增殖细胞核抗原(PCNA)表达的关系,以深入探讨P53蛋白的表达在脑星形胶质细胞瘤发生中的作用。方法:用P53蛋白抗体PAb1801和PCNA的抗体Pc10检测42例脑星形胶质细胞瘤切除标本,以阳性指数的表达方式,对两种免疫组化的结果进行定量,并对其中资料完全的35例P53蛋白和PCNA的阳性指数进行相关性分析。结果:在各级星形胶质细胞瘤中均有P53阳性表达;P53阳性指数分别为Ⅰ级14.66±13.97,Ⅱ级36.54±19.24,Ⅲ~Ⅳ级28.87±25.25;Ⅰ级和Ⅱ级组有显著差异(P<0.05)。P53和PCNA两者的阳性指数呈非常显著的正相关(r=0.512)。结论:提示P53蛋白在调控细胞增殖、促成脑星形胶质细胞瘤的生成中具有一定作用。 Objective:To clarify the correlation between the expression of P53 protein and that of PCNA,then to define further the effect of P53 expression on tumorigenesis of astrocytic tumors.Methods:42 surgical specimens of human astrocytic tumors were immunostained with a monoclonal antibody against P53 protein(PAb1801),and a monoclonal antibody against PCNA(Pc10).By counting positive index(PI),we quantitated the results of immunohistochemistry and compared the correlation between PI of P53 protein and PCNA. Results:P53 expression was observed in the tumors of all grades.The PI of P53 protein was 14.66±13.97 in grade Ⅰ, 36.54±19.24 in grade Ⅱ and 28.87±25.25 in grade Ⅲ  ̄ Ⅳ,respectively.There were significant differences in PI between grade Ⅱ and Ⅰ.We found that expression of P53 protein was related to that of PCNA,and there was a positive correlation between PI of P53 and PCNA.Conclusion:P53 palys a role in modulating cell proliferation and promoting astroglioma tumorigenesis.
