摘要
目的探讨IL-2活化特异性瘤苗联合氟尿嘧啶(5-FU)腹腔化疗,增强腹腔巨噬细胞抗肿瘤转移的效果及其免疫机制。方法建立小鼠结肠癌腹腔转移模型,用5-FU化疗后,辅助结肠癌特异性瘤苗皮下和IL-2腹腔注射,观察对小鼠腹腔巨噬细胞生物学的影响。结果IL-2联合瘤苗注射组腹腔巨噬细胞分泌IL-1、TNF-α和NO水平增高,巨噬细胞的吞噬能力和对肿瘤细胞的杀伤活性提高,显著提高腹腔巨噬细胞抗肿瘤免疫的能力。结论化疗辅助IL-2联合瘤苗免疫治疗,能优化肿瘤局部的抗肿瘤微环境,活化腹腔巨噬细胞抗肿瘤的生物学活性,对抑制结肠癌术后腹腔转移有着重要的临床意义。
Objective To investigate the immune mechanism and enhanced anti-tumor effect of abdominal cavity metastasis by 5-FU and IL-2 activated vaccine in mice.Methods The animal model of colon carcinoma metastasis in abdominal cavity were built.The tumor-bearing mice were treated with IL- 2 intra-abdominal cavity injection and CT26 colon carcinoma vaccine subcutaneous injection after 5-FU chemotherapy,the therapeutic effects and biologic activity of macrophages were observed.Results After 5-FU chemotherapy,the high level of IL-1,TNF-α and NO were excreted, the phagocytosis and cytotoxity were enhanced of macrophages in groups by using IL-2 added vaccine.Conclusion Using IL-2 and vaccine immunotherapy could enhance biologic activity of macrophages and optimize the microenvironment of anti-tumor, and inhibit tumor metastasis.
