摘要
目的考察阿魏酸哌嗪(PF)在大鼠各肠段的吸收。方法采用在体肠吸收实验,在浅麻醉状态下,分肠段考察大鼠对PF的吸收动力学。采用RP-HPLC测定循环液中药物的浓度。结果大鼠胃、十二指肠、小肠、结肠对PF均有较好的吸收,2h后的吸收百分率分别为62.04%±11.62%、70.02%±5.21%、72.08%±3.91%和12.68%±1.22%,其中十二指肠、小肠、结肠对PF的吸收速率常数分别为0.561±0.006、0.630±0.071、0.098±0.007 h-1。结论PF在大鼠胃至结肠各部位均有较好的吸收,吸收最佳部位是胃、十二指肠和小肠,在结肠部位也有少量的吸收。
OBJECTIVE To provide biological pharmaceutical basis for dosage forms design of Piperazine ferulate by studying its absorption kinetics in various intestinal segments of rat. METHODS The absorption kinetics of Piperazine ferulate from different intestinal segments were investigated using in situ perfusion method in the rats. The drug concentrations in cycling system were determined by RP - HPLC. RESULTS Piperazine ferulate was well absorbed in gastro,duodenum,intestine and colon. The absorbed rates were 62. 04% ± 11.62% ,70.02% ±5.21% ,72.08% ±3.91% and 12.68 % ±1.22%, respectively. The absorption rate constants (Ka) were 0.561 ±0.006,0. 630 ±0.071 and 0.098 ±0.007 h^ -1 in duodenum, intestine and colon. CONCLUSION Piperazine ferulate is well absorbed in gastro, duodenum, intestine and colon. The best absorption locations are gastro, duodenum and intestine; even colon can absorb piperazine ferulate too.
