摘要 为了建立从生物制品和人粪便中快速分离鉴定双歧杆菌的方法,利用双歧杆菌16S rRNA特异性扩增引物,通过PCR技术对双歧杆菌制剂和人粪便中分离菌种进行快速鉴定;同时结合革兰氏染色、穿刺实验、糖发酵实验等技术对分离的双歧杆菌的生理生化等特征进行了研究。琼脂糖电泳结果表明,利用特异性扩增引物的扩增产物在1 500 bp处出现特异性条带,革兰氏染色、糖发酵试验等也验证了分离菌种为双歧杆菌。说明利用16S rRNA的PCR方法鉴定双歧杆菌具有快速、灵敏、操作简单等优点,可以直接将双歧杆菌快速鉴定到属。 The rapid method was developed to identify Bifidobacterium from biologicals preparations and human feces. Using the special primers to amplify the 16S rDNA by PCR, the Bifidobacteriurn in Biological preparations or human feces was rapidly identified. At the same time, the physiological and biochemical characteristics of Bifidobacterium were studyed by methods such as Gram's staining, carbohydrate fermentation test, puncture test, determination of growth curve, etc. Results demonstrated that the isolated strains were Bifidobacterium. This paper showed that identification of Bifidobacteria could be simply fast completed with the PCR amplification of the 16S rDNA.
