肝干细胞分化的基因调控研究
A study of gene regulation on hepatic stem cell differentiation
摘要
目前原位肝移植是治疗各种晚期肝脏疾病的有效措施,但由于合适供体肝脏的缺乏和长期使用免疫抑制药物所导致的不良反应,大大限制了该技术的广泛应用。研究表明,肝细胞移植(hepatocyte transplantation,HCT)技术可以解决目前肝脏移植所面临的部分问题。HCT的供体来源主要包括异体肝细胞、自体肝干细胞、胚胎肝细胞、胚胎干细胞。成熟的供体肝细胞在应用于治疗时也受到组织来源的限制,并在体内难以长期增殖和维持生物学功能。晚近,具有增殖和分化潜能的肝干细胞(HSC)成为细胞移植治疗另一重要的供体来源,利用基因调控手段上调HSC分化基因表达,将可改善HSC移植治疗水平,是该领域的研究热点。
