Effect of the Metabolic Substances Produced by Common Dermatophytes on the Immune Activity of Human Keratinocytes in Vitro

摘要 目的探讨常见致病真菌代谢产物对人角质形成细胞(HKC)产生细胞因子的影响。方法红色毛癣菌、须癣毛癣菌、絮状表皮癣菌、犬小孢子菌和白念珠菌常规培养,提取培养上清液后按不同稀释度加入体外培养的HKC中,24h后观察细胞形态变化,ELISA法检测细胞上清液IL-8,IL-6及TNF-α的浓度。结果5个组均使细胞形态发生变化。在红色毛癣菌、白念珠菌、须癣毛癣菌组1∶10,犬小孢子菌组1∶1时,HKC上清液中的IL-8及IL-6均高于空白对照(P<0.01)。结论体外培养的真菌培养上清液对HKC的活性有明显的影响。浓度较高时可直接导致HKC的死亡;浓度低时可明显刺激HKC增加细胞因子的产生,其中红色毛癣菌作用最强。 Objective To observe the effects of the metabolites produced by common derrnatophytes and Candida albicans on cytokines production by human keratinocytes ( HKC ) in vitro. Methods T, rubrum, T. mentagrophyte, E. fl, occosum,M, canis and C. albicans were cultured and their supernatants were harvested, Primary human keratinocytes were cultured and treated with fugnal supernatants at different dilution (1: 1 and 1: 10) for further 24 hours, We observed the morphological changes of cells and detected the concentrations of IL-8, IL-6 and TNF-α in the cell supernatants using ELISA kits, Results Metabolites of all common fungi and C, albicans induced obvious morphological changes of keratinocytes, When the dilution of T, rubrum, T. rnentagrophytes, E, floccosum and C, albicans were at 1: 10 ,and M, canis at 1: 1 respectively,metabolites produced by the four strains induced significant elevation of IL- 8 and IL- 6 by the cells (P 〈 0.01 ). Conclusion Metabolites of dermytophytes and C, albicans influenced the biologic function of HKC in vitro. Higher concentration of the metabolites damaged HKC while they stimulated cytokine production at lower concentration.
