腺病毒介导的wt-p53基因对人瘢痕疙瘩成纤维细胞增殖的影响 被引量:7

摘要 目的 探讨野生型p53(wtp53)基因对体外培养的瘢痕疙瘩成纤维细胞(KFB)增殖的影响。方法 通过腺病毒载体将wt-p53基因转染至KFB中,用反转录-聚合酶链反应(RT-PCR)方法检测转染组与非转染组KFB中wt-p53的mRNA表达;通过间接免疫荧光法检测转染组与非转染组KFB中wt-P53蛋白表达;转染后1~6d用台盼蓝染色法计数活细胞数,并绘制生长曲线;采用流式细胞仪检测两组细胞生长周期各时相分布。结果 两组细胞中wt-p53的mRNA表达相差明显,转染组细胞中wt-P53蛋白表达明显强于非转染组;转染组细胞G0~G1期比例明显高于非转染组(P〈0.05),而G2~M期比例明显低于非转染组(P〈0.05)。结论 wt-p53基因能明显抑制体外培养的KFB增殖。 Objective To study wt-p53 gene's influence on the proliferation of keloid fibroblasts in vitro. Methods wt-p53 gene was transfeeted into keloid fibroblasts by adenovirus vector, wt-p53 mRNA was analyzed by RT-PCR; wt-p53 protein was evaluated by indirectiy immunofluorescence; The ability of proliferation of keloid fibroblasts was analyzed by cell growing curves; The cell cycle of KFB was checked by FCAS. Results The expression of wt-p53 mRNA and protein was obviously higher in the fibroblasts of the experimental group than those of control group; the rate of G0-G1 in cell cycle was higher in the fibroblasts of the experimental group than those of control group; at the same time, the rate of G2 - M was lower in fibroblasts of the experimental group than those of control group (P〈0.05). Conclusion wt-p53 gene can effectively inhibit the proliferation of keloid fibroblasts in vitro.
