摘要
目的探讨平消胶囊抑瘤乳腺癌细胞MCF-7的分子机制及其作用靶点。方法平消胶囊按照每只大鼠1.656 g/d灌胃给药,连续7 d。末次灌胃1 h后,动物乙醚吸入麻醉,心脏采血,无菌分离血清,经56℃,30min灭活处理后,再以0.22μm微孔膜过滤除菌,置-70℃保存备用。以5%、10%、15%三种不同体积分数的平消胶囊含药血清培养人乳腺癌细胞MCF-7,然后以MTT技术及基因芯片技术,体外检测平消胶囊对人乳腺癌细胞生长的抑制作用和对人肿瘤信号传导基因表达的影响。结果5%、10%、15%的平消胶囊含药血清对人乳腺癌细胞的抑瘤率分别为28.2%、29.16%、31.12%;10%平消胶囊含药血清处理乳腺癌细胞后,对人肿瘤信号传导基因芯片表达下调和上调的基因分别为34个和22个。不同体积分数的平消胶囊混悬液对人乳腺癌细胞均有一定的抑制作用,其中以15%的混悬液抑瘤率最高,但三种不同体积分数混悬液间的抑瘤率差别无统计学意义。结论平消胶囊对人乳腺癌细胞MCF-7细胞的部分信号传导基因具有显著的调节作用,这种调节作用总的趋势是有利于抑制人乳腺癌细胞MCF-7细胞生长、有利于降低其耐药性和浸润及转移潜力。
Objective To investigate the anti-cancer molecular mechanism of Pingxiao capsule, and discovery the molecular target through MTT and gene chip experiments in vitro. Methods Different density of Pingxiao capsule were cultivated with human breast carcinoma cell line MCF-7, and then detect the inhibitory action of breast cancer cell with MTT methods, and gene expression changes of signal transduction was detect with gene chips techonogy. Results The breast cencer cell inhibition rates of the three different drug concentration(5%, 10% and 15%) were 28.20//00, 29.16% and 31.12%, respectively. The numbers of the down-regulated signal transductive genes were 34 different kinds and 22 kinds of upregulated gene were detected. Conclusion The different density groups all appear to have inhibition rate to human breast cancer cell, and the 15% concentration to be the highest, but three groups does not have the obvious difference; Significant changes were observed in signal transduction expression of human breast cancer cell. The tendency is advantageous in the suppression to MCF-7 cell growth.
