摘要
目的 建立金银花主要有效成分绿原酸总量的快速比色测定方法。方法 采用酚类物质中酚羟基与三氯化铁的显色反应,对绿原酸与三氯化铁的显色反应的一些特征参数(如显色反应的条件、显色产物的吸收波长、稳定性、标准工作曲线及回收率等)进行考察与测定。结果 显色反应物的检测波长755nm;以绿原酸的吸光度和浓度得出工作曲线,回归方程y(μg/mL)=231.77x+5.4114,相关系数0.9982;绿原酸浓度0~181.0μg/mL,符合比耳定律;表观摩尔吸收系数ε=1.495×10^3L·mol^-1·cm^-1,检出限3.78μg/mL,回收率92.22%。结论 快速比色测定方法简单、快速,可望用于金银花的检测与评价。
Objective To establish the method of rapid determination of chlorogenic acid in flowers of Lonicerajaponica. Methods According to the color reaction, which was the specific color reaction occurring in FeCl3 and phenolic hydroxide group, the reacting conditions were tested, such as wavelength, stability, standard operation curve and recovery rate so on. Results 755 nm was chosen as the detective wavelength. The regression equation was Y(μg/mL)=231.77X+5. 4114, R2 =0. 9982. The molar ab- sorptive index was, 1. 495 ×10^3L·mol^-1·cm^-1. The detective limit was 3.78μg/mL and the recovery rate was 92.22%. Conclusion The simple, rapid and low-cost method can be applied to evaluate the quality of Lonicera japonica in marketing.
