摘要
以甘蓝型油莱奥罗(Oro)为受体,诸葛菜(Orychophragmusviolaceus)为供体,用花粉管通道法,在自花授粉24小时后导入诸葛菜总DNA,其后代发生了显著变异。变异率达9.8%。分析比较变异株DI-10和Oro的经济性状表明,主花序有效长度增加29.8%,第一次有效分枝数增加33.3%,有效荚果数增加176.9%,每荚粒数增加18.5%,千粒重增加44.2%,单株产量增加3.67倍;含油量增加2.42%,油酸含量增加14.33%,芥酸减至0.68%。Southren杂交表明,转化油菜(DI-10)产生优良遗传特性是诸葛菜DNA导入所致。
Exogenous total DNA extracted and purified from the donor (Orychophragmus violaceus) was microinjected into the ovary of B. napus (ORo)through the pollen tube channel 24hr after artificial pollination, and 9. 8% offsprings varied. The analyses of economic characters for varied plant (D10) showed that valid length of main inforesence increased by 29. 8%;No. of lst valid branches increased by 33. 3 % ; no. of valid pod increased by 176. 9% ; seeds. per pod increased by 18. 5 % ;1000-grain weight increased by 44. 2% (g) ; yield /plant increased by 3. 67 times. and the oil contents increased by 2.42%. Meawhile, the contents of oleic acid increased by 14. 33% ; erucic acid decreased to 0. 68%. Southren blot showed that orychophrgmus violaceus DNA's intervention were induced to varied B. napus, which proved that O. vidaceus DNA was superior character recipient.
