复合抗氧化剂对大鼠肝癌阿霉素化疗外周血的保护作用 被引量:1

摘要 目的观察大鼠肝癌模型腹腔阿霉素化疗引起的外周血的变化以及复合抗氧化剂的保护作用。方法采用Walker-256肿瘤细胞株接种大鼠皮下,得到实体瘤,将实体瘤分割成小块植入大鼠肝脏,制成大鼠移植性肝癌模型,将肝癌模型大鼠随机分成3组,分别为模型组、单纯化疗组和抗氧化剂保护组,同时选取经手术但未接种肿瘤的大鼠作为阴性对照组。抗氧化剂保护组每日给予复合抗氧化剂灌胃,单纯化疗组和抗氧化剂保护组大鼠每2天进行1次腹腔阿霉素化疗,剂量为2mg/kg体重,第8周末处死大鼠,取抗凝全血测定外周血各项指标。结果与阴性组和模型组相比较,单纯化疗组的白细胞数(WBC)、淋巴细胞百分比(LYM%)、淋巴细胞数(LYM)、红细胞数(RBC)、红细胞压积(HCT)、血红蛋白(HGB)、平均血小板体积(MPV)显著降低(P<0.05),单核细胞百分比(MON%)、中性粒细胞百分比(GRA%)、单核细胞数(MON)、中性粒细胞数(GRA)、平均红细胞体积(MCV)、红细胞分布活力(RDW)、平均红细胞血红蛋白含量(MCH)、血小板数(PLT)、血小板分布活力(PDW)显著升高,而抗氧化剂保护组与单纯化疗组相比呈现出了明显的拮抗保护作用。结论大鼠肝癌腹腔阿霉素化疗可以造成外周血损伤,引起血象的明显改变,而复合抗氧化剂可以起到有效地保护作用。 [Objective] To investigate the injury to peripheral blood in rats with liver embedded-tumor treated by adriamycin and the protective effect of multiple antioxidants. [Methods] The tumor cells (Walker-256) were injected into the rat to create the entity tumor. The entity tumour removed from rat was cut into small pieces for being embedded into the rats' livers. The rats with entity tumor were randomly divided into three groups which were tumor group, chemotherapy group and protective group. The protective group was given anfioxidants compound by gavage and other two groups were given the same dosage water. The chemotherapy group and the protective group were given adriamycin by intraperitoneal injection with the dosage of 2 mg/kg body weight every two days. At the eighth week end, all of the animals were killed to measure the changes of peripheral blood components. [Results] Compared with the control group and the tumor group, WBC, LYM%, LYM, RBC, HCT, HGB, MPV of peripheral blood in chemotherapy group decreased significantly (P 〈0.05), and MON%, GRA%, MON, GRA, MCV, RDW, MCH, PLT, PDW of peripheral blood increased significantly (P 〈0.05), on which the multiple antioxidants showed remarkable antagonistic effect. [Conclusion] The chemotherapy of the hver tumor with adriamycin can cause injury to peripheral blood in rats, on which the multiple antioxidants have effective protection.
