摘要
目的:观察不同浓度肿瘤坏死因子-α(TNF-α)在不同时间对人血管内皮细胞(Endothelial cell ofvessels,ECV)中细胞间粘附分子-1(ICAM-1)基因表达的影响及中药尿酸利仙(含药血清)的干预作用。方法:①体外培养人血管内皮细胞,分别用不同浓度(12.5、25、50μg/mL)的TNF-α刺激,并用25μg/mL的TNF-α分别孵育细胞8、16、24、48h;②用25μg/mL的TNF-α孵育细胞16h,同时用含不同浓度(2.5%、5%、10%)尿酸利仙含药血清的培养液进行干预。用逆转录-聚合酶链式反应(RT-PCR)技术测定血管内皮细胞ICAM-1的mRNA表达。结果:①低、中、高浓度TNF-α刺激均可显著上调内皮细胞ICAM-1 mRNA的表达,且体现了一定的浓度梯度关系,但刺激组间无显著性差异。②25μg/mL的TNF-α在8h、16 h、24 h和48 h均可显著上调内皮细胞ICAM-1 mRNA的表达,各干预组间无显著性差异。③低、中、高浓度的尿酸利仙含药血清干预均可显著降低ICAM-1 mRNA的表达(P≤0.01),并以中浓度尿酸利仙含药血清作用最为显著。结论:TNF-α能显著上调内皮细胞表面黏附分子ICAM-1的mRNA表达,而尿酸利仙含药血清抑制病理条件下ICAM-1基因水平的过量表达,可能是其减轻TNF-α诱导的人血管内皮细胞炎症损伤的机制之一。
Objective: To investigate the effect of the herbal compound (NiaoSuanLiXian) on the expression of intercellular adhesion molecule ( ICAM - 1 ) mRNA in vascular endothelial ceils induced by TNF -α. Methods Cultured the vascular endothelial cells, and stimulated with TNF -α. At the same time, the ceils were co - incubated with the serum including the herbal compound. The expression of ICAM - 1 mBNA in the vascular endothelial cells was detected by RT - PCR. Results The expression of ICAM - 1 mRNA in the vascular endothelial cells induced by TNF -α increased significantly than that in the control cells. 2.5% , 5% and 10% of the serum including NiaoSuanLiXian could all significantly down - regulate the expression of ICAM - 1 on levels of mRNA. Conclusion The role of down - regulating the expression of ICAM - 1 mRNA of NiaoSuanLiXian in the vascular endothelial cells could be One of the mechanism of reducing the inflammatory injure and preventing the arteriosclerosis.
