高分压氧处理对体外培养大鼠脾淋巴细胞增殖及杀伤能力的影响

摘要 目的观察大鼠体外脾淋巴细胞经不同压力—时程的高分压氧处理后淋巴细胞增殖及细胞毒性T淋巴细胞(CTL)杀伤能力的变化。方法分离成年大鼠脾淋巴细胞,置于高压细胞培养舱内,以不同压力—时程的高分压氧(含CO25 kPa)处理,W ST-8比色法测定经刀豆球蛋白(ConA)刺激后淋巴细胞的增殖率和对肿瘤细胞株P815的杀伤能力。以常压空气(含5%CO2)处理作为对照组。结果一定压力—时程的高分压氧处理对体外培养的大鼠脾淋巴细胞增殖及杀伤能力具有促进作用,但随暴露剂量的增加则逐渐表现为抑制效应。结论高分压氧对离体大鼠脾淋巴细胞增殖及杀伤能力存在一个从适度暴露促进功能发挥至过量暴露抑制功能发挥的过程。 Objective To observe the effects of hyperbaric oxygen on the proliferation rate and total cytotoxicity T lymphocyte (CTL) activity of rat splenic lymphocytes. Methods Splenic lymphocytes of adult rats were separated and treated in a hyperbaric cell-culture chamber at the different pressure-duration of O2 ( containing 5 kPa CO2 ). Lymphocyte proliferation rate and total CTL activity stimulated by ConA were determined with WST-8 colorimetry. Results Lymphocyte proliferation rate and total CTL activity were promoted after some certain doses (pressure-duration) of hyperbaric O2 treatment but inhibited at higher doses. Conclusion Hyperbaric O2 has dual effects on the functions of rat splenic lymphocytes that proper exposure promotes but excessive exposure inhibits them.
