水稻 c DNA文库的构建以及 1 0 0种随机挑取克隆部分结构的分析 被引量:4

摘要 以水稻 (Oryza sativa)“广陆矮 4号”黄化苗为材料 ,用 λZAP 为载体构建了 c DNA文库。对随机挑取的 1 0 0个 c DNA进行部分顺序测定后 ,与水稻和其它植物已知的基因结构作同源性比较分析 ,表明1 3%的 c DNA克隆可以确定功能 ,1 2 %的 c DNA克隆与其它植物中未知功能的基因片段有同源性 ,其余75%的克隆则表现出极少相似性或无任何同源性 ,从中可能发现新的基因。结果表明这个 c DNA文库适宜于进行大量顺序分析。由此将会获得更多功能性基因的信息。 A cDNA library of rice (Oryza sativa ssp. indica cv. “Guanglu' ai 4”) etiolated shoot was constructed using Lambda ZAPⅡ vector. After analysing the partial sequences of 100 randomly selected clones and database comparisons to rice and other plants, 13% cDNA clones could be identified and 12% cDNAs had high degree of sequence similarity to partial sequence from rice or other species, whose function is still unknown. The remaining 75% cDNAs showed little or no similarity to genes in the database and might represent novel genes. It demonstrates the suitability of this library for large scale sequencing from which more information of functional genes will result.
