大鼠脑出血后氧化还原因子-1表达与细胞凋亡相关性的研究

摘要 目的探讨脑出血(ICH)后血肿周围组织中氧化还原因子1(Ref1)表达与细胞凋亡的关系。方法应用立体定向技术,将SD大鼠自体不凝血50μl注入其尾状核区制备ICH模型,分别在不同时间点断头取脑,连续切片作Ref1免疫组化染色和原位末端标记(TUNEL)染色;观察各时间点Ref1表达及TUNEL阳性细胞数,并进行相关性分析。结果ICH后血肿周围组织中TUNEL阳性细胞与Ref1表达呈负相关(r=-0.745,P<0.05),且Ref1表达下降谷底时间明显早于细胞凋亡高峰的时间。结论ICH后血肿周围组织中细胞凋亡增加与Ref1表达减少可能有关。 Objective To investigate the correlation between the expression of redox factor-1 (Ref-1) and neuronal apoptosis around hematoma after experimental intracerebral hemorrhage ( ICH ) in rats. Methods ICH model of rat was induced using stereotaetic infusion antilogous blood 50μl into the caudate nucleus. The male animals were randomly divided into sham operation group, normal control group and hemorrhage group. TUNEL and immunohistochemistry methods were used to detect apoptosis and the expression of Ref-1 in cerebral tissues at different time, respectively. Results The expression of Ref-1 was negatively correlated with neuronal apoptosis around hematoma after ICH ( r = - 0. 745, P 〈 0. 05 ). The descending of Ref-1 expression preceded the peaking of neuronal apoptosis. Conclusion Neuronal apoptosis after ICH is associated with the activation of Ref-1.
