摘要
目的采用同种移植技术建立Wistar大鼠肝细胞癌模型,并初步探讨其MRI表现。材料与方法将CBRH-7919大鼠肝癌细胞注入免疫抑制的2只6周龄Wistar大鼠皮下,14d后将生成的瘤块移植到免疫抑制的33只实验组大鼠肝内。肝内瘤块移植后第5、10、15、20、25d分别取相应时间点实验组全部存活大鼠及随机抽取的1只对照组大鼠行磁共振成像并进行影像分析,然后处死动物,取实验组肝内肿块及对照组肝组织做常规病理检查。结果24只大鼠造模成功,病理证实为肝细胞癌,成功率为72.73%。接种10d后19只大鼠的肝内瘤灶均被MR平扫检出,呈稍长或等、稍长混杂T1信号,均匀或不均匀稍长、长T2信号。结论通过免疫抑制同种移植技术可建立Wistar大鼠CBRH-7919原位肝细胞癌模型。该种建模方法操作简便,易于重复,移植成功率高,实验周期短,肿瘤MRI表现与人类肝细胞癌信号特点相似,是一种较理想的适合于肝癌影像学评价研究的动物模型。
Objective To establish an homogeneous implantable Wistar rat model of hepatoceilular carcinoma and discuss its magnetic resonance imaging features. Materials and Methods Subcutaneous inoculation of CBRH 7919 rat liver cancer ceils were performed in two six weeks old male Wistar rats, which immune system were inhibited. After 14 days' growth, the tumor fragments were transplanted into left liver of 33 male immune suppressed rats in experimental group. 1 rat of the control group and all survival rats of experimental group in different developmental stages were performed with plain MR scan after 5,10, 15,20,25 days of Homogeneous implantation respectively. The MRI findings were analyzed, then the rats were killed and cancerous tissues of different developmental stages and liver tissues of the control group were examined pathologically, Results Homogeneous implantable rat model of bepatocellular carcinoma were established successfully in 24 rats. The pathological examination showed that the tumors were hepatocellular carcinoma, the achievement ratio was 72.73%. MRI showed all liver tumors of 19 rats 10 days after implantation. The tumors were isointensity or hypeintensity on T1WI and homogeneous or inhomogeneous hyperintensity on T2WI. Conclusion A rat liver cancer model can be established by immune suppression homogeneous implantation. Convenient operation and good reproducibility, high achievement ratio, short experimental periodicity and similar MRI characteristics to human HCC make this tumor model highly suitable for the evaluation study of HCC by radiology.
