高效液相色谱法测定人血浆中盐酸环丙沙星的方法学研究 被引量:5

摘要 目的 建立血浆中盐酸环丙沙星含量的RP—HPLC测定方法。方法 采用Diamonsil C18色谱柱(200mm×4.6mm,5μm);乙腈-20mmol·L^-1磷酸氢二钾/磷酸二氢钾缓冲溶液(用磷酸调pH至2.83)=25:75为流动相,用前经0.45μm的滤膜过滤,超声脱气;流速为1.0mL·min^-1;荧光检测波长为295/440nm;柱温为35℃,进样量20μL。结果 此方法最低检测浓度为5ng·mL^-1,线性范围为10~4000ng·mL^-1,方法平均回收率在99.11%~103.80%,日内及日间精密度的相对标准差分别〈2%和5%。结论 该方法简单、快速,灵敏度高,重复性好。 OBJECTIVE To determine eiprofloxacin hydrochloride in the human plasma by reversed phase high performance liquid chromatography. METHODS Ciprofloxaein hydroehloride was successfully separated using a Diamonsil C18 column (200 mm×4.6mm,5μm). The mobile phase consisted of aeetonitrile-20 mmol ·L^-1 dibasicpotassium phosphate and monobasicpotassium phosphate buffer solution (25 : 75) (adjusted to pH 2.83 using phosphoric acid) and was degassed ultrasonically before use. Each component of the mobile phase was filtered through a 0. 45 μm membrane. The column temperature was 35 ℃ and the flow rate was 1.0 mL ·mL^-1. Injection volume was 20 μL. The peak of eiprofloxacin hydroehloride was monitored with fluorescence detector (290/440 nm). RESULTS The detection limit was 5 ng ·mL^-1. The linear range was 10~4 000 ng ·mL^-1. The average recovery was 99.11%~103. 80%. RSD of intra-day and inter-day was less than 2% and 5%, respectively. CONCLUSIONS The method is simple, rapid, sensitive and producible.
