摘要
观察人肝癌细胞中特异性环氧合酶-2(Cyclooxygenase-2,COX-2)与MDR/P—gp表达变化,探讨二者在Celecoxib诱导细胞凋亡中的意义。采用免疫组化法、流式细胞技术以及RT-PCR观察Celecoxib诱导肝癌细胞凋亡过程中COX-2与MDR/P—gP表达的变化。Celecoxib诱导肝癌细胞呈时间、剂量依赖性;细胞周期分布改变,G0/G1期细胞比例增加;COX-2与MDR/P—gp表达减弱,但非线性相关。Celecoxib可能通过降低COX-2、MDR/P—gp表达,影响细胞周期分布,发挥抗肿瘤作用。
To evaluate the expression of Cyclooxygenase -2 ( Cox -2) and MDR/P - gp during apoptosis of hepatocarcinoma cells induced by celecoxib. The expressions of Cox -2 and MDR/P-gp were evaluated by immunohistochemistry, flow cytometry and RT - PCR. Apoptosis induced by celecoxib was time - and concentration - dependent. The percentage of G0/G1 phase cells increased compared with the controls ( P 〈 0.01 ) and other groups. COX - 2 and MDR/P - gp were dowm- regulated. But there was no relationship between them. Above findings suggested that celecoxib might has anti carcinoma effect through decreasing the expression of Cox - 2 and MDR/P - gp and changing cell life cycle.
