摘要
本文对人血清中白蛋白的鲁米诺-K3Fe(CN)6化学发光反应体系进行了较系统的研究。考察了流速、发光试剂浓度、PH值以及增敏剂等对人血清白蛋白分析检测的影响。在优化的实验条件下,测定人血清中白蛋白的线性范围为1.2×10^-11~4.4×10^-8mol/L,R^2=0.9942;方法的检出限(S/N=3)为1.2×10^-11mol/L;回收率范围为90.75%~103.5%,本法对2.9×10^-8mol/L的标准样品平行重复12次测定,其相对标准偏差为3.5%。
A new CL reaction of HSA-Luminol-K3Fe(CN)6 was studied by flow injection method. The factors of influencing CL intensity and the enhancement of the surfactants were investigated. A new method based on the CL reaction for the determination of HSA has been established by optimizing the experimental conditions. The linear range was 1.2×10^-11-4.4× 10^-8 mol/L,R^2=0. 9942. The detection limit for HSA was 1.2× 10^-11 mol/L(S/N= 3) . The recovery rate was at 100 ± 10%. For a standard sample (c=2.9×10^-8mol/L),the RSD was lower than 5%(n=12).
