摘要
目的 建立反相高效液相色谱(RP-HPLC)法测定复方大黄颗粒中大黄素和大黄酚的含量。方法 Alltima C18色谱柱(5μm,150mm×4.6mm);柱温:30℃;流动相:甲醇-0.1%磷酸(87:13);流速:1ml/min;检测波长:254nm。结果 大黄素的线性范围为0.096~0.576μg(r=0.9999),平均回收率为99.29%,RSD为0.63%;大黄酚的线性范围为0.0784~0.4704μg(r=0.9998),平均回收率为100.08%,RSD为1.52%。结论 该方法灵敏度高、操作简便、重现性好、效率高。可作为样品的检测方法。
Objective To develop an RP-HPLC method for determination of emodin and chrysophanol in Compound Rhubard Granula. Methods The samples were determined by RP-HPLC with a Alhima C18 column(5μm,150mm×4.6mm) and UV detector at 254 nm. The mobile phase was methonal: 0.1% H3 PO4 (87: 13) at a flow rate of 1.0 mL/min. The column temperature was 30℃. Results The calibration curve of emodin was linear over the ranges of 0.096μg ~ 0. 576 μg ( r = 0. 999 9 ), average recovery was 99.29 ,RSD was 0.63% ;The calibration curve of chrysophanol was linear over the ranges of 0. 078 4 ~ 0. 470 4 μg (r =0. 999 8), average recovery was 100.08%, RSDwas 1.52%. Conclusion This method is simple, reliable, efficient, and it provides a reliable method for determination of emodin and chrysophanol in Compound Rhubard Granula.
