川芎嗪对大鼠肠缺血再灌注肝损伤保护作用的实验研究 被引量:1

Protective Effects of Tetramethylpyrazine on Hepatic Damage Induced by Intestinal Ischemia-reperfusion in Rats

摘要 目的探讨川芎嗪对大鼠肠缺血再灌注肝损伤的保护作用。方法选用健康W istar大鼠24只,随机分成3组(每组8只):假手术对照组、缺血再灌注组、川芎嗪治疗组。夹闭大鼠肠系膜上动脉(SMA)60 m in造成缺血,再灌注2 h后取出肝组织制成匀浆,测定超氧化物歧化酶(SOD)、谷胱甘肽过氧化酶(GSH-PX)、丙二醛(MDA)、Ca2+-Mg2+-ATP酶的含量及肝形态细胞学变化。结果川芎嗪治疗组肝组织MDA含量明显低于缺血再灌注组(P<0.01),SOD,GSH-PX,Ca2+-Mg2+-ATP酶的含量均明显高于缺血再灌注组(P<0.01),肝细胞形态学异常变化明显减轻。结论川芎嗪能减少大鼠脂质过氧化,改善ATPase功能,减轻大鼠肠缺血再灌注所致肝脏损伤。 Objective To explore the protective effects of tetramethylpyrazine on hepatic damage induced by intestinal ischemiareperfusion in rats. Methods Divide the rats randomly into three groups: (1)sham operated group, (2)untreated ischemia-reperfution(I/R) group, (3)pretreatment with tetramethylpyrazine before I/R(Te). Superior mesenteric artery was clipped for 60 min to cause ischimia, and then unclipped to make reperfusion for 2 hours after reperfusion, the liver tissues were gathered and homogenized respectively, and the contents of SOD, GSH-PX, MDA and ATPase, and morphological changes of hepatocytes were assayed. Results The contents of MDA in liver in Te groups were less than that in I/R group (P 〈0.01 ) ,the contents of SOD, GSH-PX and Ca^2+ ,-Mg^2+ ,-ATPase in Te groups were higher than those in I/R group( P 〈 0.01 ), and the abnormal morphological changes of hepatocytes diminished remarkably. Conclusion Tetramethylpyrazine can reduce lipid peroxidation, improve ATPase activity and decrease the injury of liver tissue in the rats after intestinal ischemia-repeffusion.
