摘要
目的探讨c-FLIP和TGF-β1在喉鳞癌组织中的表达与肿瘤的病理学分级和患者临床分期及颈淋巴结转移之间的相关性。方法应用RT-PCR法检测30例喉鳞癌患者肿瘤组织及喉部正常组织中c-FLIP mRNA的表达情况,同时应用免疫组化法对实验组及对照组中c-FLIP和TGF-β1蛋白的表达情况进行了检测。结果①实验组中,c-FLIP mRNA表达阳性者有18例,而对照组中仅3例表达阳性。②c-FLIP、TGF-β1在实验组中的平均阳性细胞表达率分别为44.87%和48.56%,而在对照组中此相应值分别为8.63%和8.25%。③两因子在喉鳞癌组织中的表达水平均与患者临床分期和肿瘤病理学分级成正相关,且它们在伴颈淋巴结转移组中的表达水平均明显高于非转移组。结论①c-FLIP、TGF-β1在喉鳞癌组织中高表达而在喉部正常组织中低表达。②c-FLIP和TGF-β1两因子在喉鳞癌组织中的表达水平与喉鳞癌患者的临床病理参数密切相关。③c-FLIP和TGF-β1可能共同参与了喉鳞癌的发生、发展。
Objective To investigate the correlations between expression of c-FLIP and TGF-β1 in larynx squamous carcinoma tissue and pathological grading of the tumor, clinical staging of patients, and cervical lymph node metastasis. Methods RT-PCR was employed to measure the expression of c-FLIP mRNA in tumor tissue and normal larynx tissue. Immunohistochemistry was employed to measure the related expression of c-FLIP and TGF-β1 in the two kinds of tissue,and the correlations between expression of the two cytokines and some clinicopathological indexes of the tumor were observed. Results There were 18 samples which expressed c- FLIP mRNA in the experimental group. There were only 3 samples which expressed c-FLIPmRNA in the control group. The positive rates of c-FLIP and TGF-β1 were 44.87% and 48.56% in the experimental group,respectively. Positive rates of the two cytokines were 8.63 % and 8.25 % in the control group, respectively. The expression of the two cytokines was significantly associated with clinical staging,pathological grading and cervical lymph node metastasis. Conclusion The expression of c-FLIP and TGF-β1 is significantly higher in tissue of Larynx squamous carcinoma than those in normal larynx tissue. The expression of the two cytokines in the tissue of larynx squamous carcinoma is significantly correlated with clinicopathological indexes of the tumor. The two cytokines might participate in occurrence and development of larynx squamous carcinoma together.
